Abstract:
This work includes the construction of an experimental chamber to implement total internal reflection fluorescence microscopy (TIR-FM) on an existing set-up. Furthermore, a frequency-doubled Nd:YAG laser was tested concerning its applicability as the excitation source in forthcoming experiments. The constructed chamber allowed to generate the evanescent field needed for TIR-FM. The advantages and drawbacks of its utilisation compared to confocal imaging are discussed.
